Title: Late-Stage Functionalization for the Expansion of Chemical Space in Bioactive Compounds

Poster No.: 3P-32

Presentation Summary
Lead optimization in drug discovery often involves structurally complex compounds including multiple heteroaromatic rings and diverse functional groups. Applying Late-Stage Functionalization (LSF), in which functional groups are introduced or transformed at a late stage of synthesis, to such compounds presents challenges in predicting reactivity and site selectivity, as well as in separating and identifying the resulting products.
In this presentation, we will showcase successful examples in which marketed drugs were used as model substrates to generate structurally diverse derivatives with different substitution patterns and functional groups through LSF.
We will also discuss the practical challenges associated with incorporating LSF into the drug discovery workflow and our efforts to address them.
