Title: Ameliorative effects of combination therapy with Resmetirom, Tirzepatide, and Roflumilast on a GAN diet-MASH mouse model

No: 1368

Presentation Summary
Recently, THR-β and GLP-1R agonists have been approved for the treatment of metabolic dysfunction–associated steatohepatitis (MASH). As these agents exert their effects through distinct mechanisms of action, combination therapy is expected to provide enhanced therapeutic benefits.
In this study, we evaluated dual and triple combination therapies in GAN diet-induced MASH mouse model. The tested agents included Resmetirom (THR-β agonist), Tirzepatide (GLP-1R/GIPR agonist), and Roflumilast (PDE4 inhibitor), which has a mechanism distinct from those of the other two compounds and potential therapeutic utility in MASH. Compared with vehicle-treated mice, each monotherapy regimen significantly improved or tended to improve liver injury markers, hepatic steatosis and NAFLD activity score (NAS). Furthermore, dual combination treatments provided greater improvements in these parameters than each monotherapy regimen. In this presentation, we will discuss the potential of combination therapies utilizing agents with different mechanisms of action as a promising strategy for MASH treatment.

Axcelead’s Soulution
We can offer various MASH model mice, including GAN diet-fed mice and Western diet-fed MC4R knockout mice, which mimic human MASH pathology accompanied by obesity and insulin resistance. We can non-invasively evaluate body composition, energy expenditure and respiratory quotient using EchoMRI and Oxymax system. In addition, we can also perform pathological analyses, omics analyses and others in one platform. Please feel free to contact us for research on MASH or other diseases.
